We used this dye to monitor cell proliferation via flow cytometry of in vitro activated T cells. We were interested in assessing early proliferation within 48hrs upon activation. This dye is quite reliable for monitoring at least six to seven divisions but it has drawbacks compared to the newer dyes out there.

The Bottom Line

Do not use CFSE. It's better to switch to other alternative proliferation dyes since they are more consistent and does not induce cell loss or viability issues.
