Extraction of high-quality plasmid DNA is a crucial step to any research lab that works with mammalian cells to model disease pathogenesis. Here, I utilized the new PureLink Fast Endotoxin Maxi Kit from Thermo Fisher, which is definitely a product that is very easy to use and extremely convenient.

200 mL culture - high copy plasmid
The kits offers a vacuum pump that makes the extraction easy. Additionally, the purification columns contain two adapters that allow easy loading of the 50 mL solution (3 buffers), and a very convenient spin column that can fit in a 1.5 mL tube for easy removal of excess wash buffer. Elution can then be done in any standard 1.5 mL eppendorff tube with a table-top centrifuge. Yield was as expected and the procedure took less than 30 minutes from the re-suspension step of the pellet.
Great for multiplexing - definetly can do the whole procedure in a 50 mL falcon tube

Expensive kit, but worth it
Great kit for fast maxi prep extraction. Plus it's endotoxin free!!
