I study CD8 T cell differentiation in both acute and chronic viral infection. PD-1 is a marker of CD8 T cell activation and exhaustion and thus I include PD-1 on almost all of my flow cytometry panels. It's an advantage to be able to detect PD-1 on multiple channels for flexibility in designing flow panels.

Results Summary

PBMCs were isolated from virus-infected mice 8 days after infection and purified by a histopaque gradient. PD-1 expression is used to identify activated/exhausted CD8 T cells; PD-1 should be undetectable on naive CD8 T cells. Picture shown is gated on CD8 T cells, with CD44 (x) vs PD-1 BV786 (y). PD-1 is only expressed on activated cells, which are marked by expression of CD44. As you can see, this antibody clearly detects PD-1 expression on the CD44+ CD8 T cells, while CD44- cells are PD-1-. I wanted to post this review as there is another review of this product that did not detect PD-1 on their in vitro simulated T cells. Our lab uses this antibody routinely for detection of PD-1 from primary cells, with great results. We stain in 2% FBS/2mM EDTA in PBS, with a 1:100 dilution of the antibody for 30 minutes.
