Aim: precipitate and immuno-probe myc-tagged proteinMaterials: For IP: 500ug cell lysate of 293T cells transfected with myc-tagged expression plasmid. For WB: 20ug cell lysate of 293T cells transfected with myc-tagged expression plasmid. Primary Ab: mouse mAb against myc tag (0.5ug/ul).Methods: 20ug cell lysate and 20ul precipitated protein were separated on a 4-15% gradient SDS-PAGE gel and transferred onto a PVDF membrane. After 2 hour blocking, the membrane was probed with anti-myc Ab. Results: As shown in the figure, in the Input group, the myc-tagged protein was detected without any visible non-specific bands. In the IP group, the detection of the precipitated myc-tagged protein was clearly observed. Conclusion: this myc antibody can be used in WB and IP assays.
