HL-60 cells are able to differentiate to neutrophils after all trans retinoic acid (RA) treatment. RARalpha expression level was assayed using Anti-Retinoic Acid Receptor alpha antibody [EP1823Y] (ab76074). Cell culture volumes containing two million cells (untreated vs. RA treated) were harvested, fixed for 10 min with 2% paraformaldehyde, and permeabilized with ice cold methanol. After 3 washes with PBS, the cells were incubated with Anti-Retinoic Acid Receptor alpha antibody [EP1823Y] (ab76074) that we previously labeled with the ThermoFisher Alexa Fluor® 488 dye. According to the Abcam datasheet, the Anti-Retinoic Acid Receptor alpha antibody [EP1823Y] (ab76074) is tested for flow cytometry. We also had good results with it.
Flow Cytometry

Cells were incubated over night , 4C, in 200µl PBS with 2.5µl RARalpha antibody

Instrument: BD FACS ARIA III SORP. The Alexa 488 emission from 488 nm excitation was collected through a 505 longpass dichroic and 515/20 bandpass filter (channel 488–1).
Anti-Retinoic Acid Receptor alpha antibody [EP1823Y] (ab76074) allows RAR alpha detection with good resolution.
