Company:

Product Name:

Anti-CFTR Antibody, clone M3A7

Catalog Number:

05-583

Cell lysates of CFBE cells expressing CFTR was resolved by SDS-PAGE followed by western blotting. There will be two distinct bands - one at ~ 140 kDa (band B; core glycosylated protein present in the ER) and another at ~ 170 kDa (band C; processed by Golgi-localized glycosylation enzymes). In the case of wild type CFTR, the majority of the protein is in the form of band C while in the case of mutant F508del-CFTR the major form is band B. Sometimes a lower molecular weight band, termed band A is also observed that probably corresponds to non-glycosylated CFTR.

Experimental Design and Results Summary

Applications

Western Blot

Sample

CFBE cell lysates expression Wt-CFTR and F508del-CFTR

Primary Incubation

1: 1000 dilution, Overnight at 4°C

Blocking Agent

5% non-fat dry milk 1hr

Secondary Incubation

Incubate membrane with Anti-mouse IgG, Peroxidase-linked Antibody (401215-MerckMillipore, 1:8000 diluted) in blocking buffer with gentle agitation for 1 hr at room temperature

Tertiary Incubation

None

Detection

Added Luminata Crescendo Western HRP substrate (WBLUR0100) on top of the blot for 3-5 min, wiped off excess solution from the blot and wrapped securely in a plastic wrap. Exposed the blot to x-ray film for 30 seconds -5 min.
