Total RNA can be extracted from whole blood using the PAXgene Blood RNA kit. Whole blood is collected from an individual into a PAXgene Blood RNA tube. The kit is phenol/chloroform free. It is a silica membrane based protocol.

Protocol Overview

Initially, cells are pelleted, washed multiple times and then protein is degraded. Samples are homogenized using the reagent provided in the kit, washed and DNAse treated. Then washed again multiple times and finally eluted using the buffer provided in the kit. The concentration and purity are determined using the Nanodrop or bioanalyzer.

Tips

Use a syringe with needle to homogenise the samples.

Results Summary

Good total RNA yield is derived from the extraction process but the purity is generally low. Good kit in the absence of alternative methods.
