Our lab performs a ton of western blots and we recently needed to blot for alpha-tubulin. We stumbled upon this antibody after borrowing an aliquot from another lab, and have since purchased it for ourselves.

Primary Incubation

1:1000 Dilution Overnight in 5% BSA in 1X TBS-T at 4C

Blocking Agent

5% Milk in 1X TBS-T

Secondary Incubation

1:5000 dilution of HRP-conjugated Mouse secondary in 5% Milk in TBS-T for 1 hour at RT

Tertiary Incubation

None

Detection

ECL --> Autoradiography Film via HRP-Conjugate Secondary Antibody

Results Summary

We get very clean results with this antibody. It comes up well on film, works with low protein levels, and lasts for a while if your lab is the type to reuse antibodies. We're pleased with the results.

While this antibody does cost a good deal more than most, it's worth it. Our blots look great and we didn't waste any time having to optimize, which is great. I highly recommend this antibody to anyone blotting for a-tubulin.
