Cre transgenes are driven by specific gene regulatory sequences, in our case, under the Mouse Insulin Promoter. The Cre-ERT2 is only translocated to the nucleus after following treatment with Tamoxifen. In this case, we gave transgenic mice 3 doses of Tamoxifen by oral gavage 3 times over 5 days. The pancreas was harvested, fixed in 4%PFA and embedded in OCT the day after the last Tamoxifen treatment.
