Company:

Invitrogen (Life Technologies)

Product Name:

Cell trace violet

Catalog Number:

C34557

We adoptively transfer T cells into recipient mice and then measure proliferation. Cell trace violet has been a great marker for proliferation. We use cell trace violet over CFSE because there is no bleed over into channels such as PE which are needed for other markers.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Murine T cells

Primary Incubation

1:1000 for 20 minutes at 37 deg C in the dark

Blocking Agent

NA

Secondary Incubation

NA

Tertiary Incubation

NA

Detection

Canto II flow cytometer

Results Summary

Great staining could still be detected even 33 days after initial staining and transfer into recipient animals. Clear cell divisions, no bleed over into other commonly used channels, less cell toxicity than what is seen with CFSE. In the sample image, antigen specific Thy1.1+ cells were stained with CTV, along with a Thy1.1neg CTV-stained antigen non-specific cotransfer which did not proliferate.

Summary

The Good

Great staining, minimal toxicity

The Bad

Extra staining step because it works best at 37 deg C

The Bottom Line

Much better staining than CFSE.
