This antibody was used to perform immunofluorescence microscopy on HeLa cells transfected with an HA-containing construct. Ab9111 was used in parallel with mouse anti-HA clone HA-7 (Sigma). Ab9111 showed moderate to strong immunoreactivity that overlapped with HA-7 staining. Background levels were acceptable, but not absent. This antibody is a good choice for multicolor microscopy when non-mouse host species antibodies are needed.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

HeLa cells transfected with HA containing construct

Primary Incubation

ab9111 and HA-7 incubated 1 hour in PBS 1% + normal goat serum +0.1% Triton X-100 at a dilution of 1:500

Blocking Agent

PBS 1% + normal goat serum +0.1% Triton X-100

Secondary Incubation

Goat anti-Rat alexa 568 and goat anti-mouse alexa 488 (Life Technologies) at a dilution of 1:500 for 1 hour

Tertiary Incubation

NA

Detection

Epifluorescence microscopy, Nikon 40X objective

Results Summary

This antibody worked well in this application. The staining was strong to moderate when compared to HA-7(Sigma) or 3F10 (Roche) and provides a viable option for non-murine antibodies.

Summary

The Good

Non-mouse, non-rat option for IF microscopy

The Bad

Not quite as strong of staining compared to HA-7 or 3F10

The Bottom Line

If you need a chicken antibody to HA, ab9111 works well. If you can use a rat anti-HA antibody I would suggest clone 3F10 from Roche.
