Company:

Santa Cruz Biotechnology

Product Name:

Anti-tyrosine hydroxylase rabbit pAB

Catalog Number:

sc-14007

This antibody was used to look at the amount of tyrosine hydroxylase protein in brain homogenates of MPTP-intoxicated mice. Midbrain containing the SN and striatal areas of the brain of mice were isolated, homogenized, and purified. 40 ug total protein was loaded and ran on 4_20% gels. Gels were transferred to PVDF membranes for incubation with Th antibody and HRP-conjugated secondary.

1:200 in 5% dry milk/PBST overnight at 4 degrees

Blocking Agent

5% dry milk/PBST for 45 - 60 minutes at room temp

Secondary Incubation

60 minutes with HRP-conjugated secondary antibody (1:20,000) in 5% dry milk/PBST

Tertiary Incubation

NA

Detection

Femto Chemiluminescent

Results Summary

Got very good signal even with a relatively short development time of the membrane.

Summary

The Good

Great signal, no non-specific bands, can re-use diluted primary antibody 2 or 3 times

The Bad

Need to use relatively high concentration.

The Bottom Line

Works great and consistently on brain homogenate once proper dilution is determined.
