I was looking for a way to speed up my western blots since I run many gels per week while doing a whole lot of other experiments. I gave this machine (is it really a machine?) a try and was surprised at how neat the membrane turned out. The most amazing thing is that it neither uses any power nor vacuum suction. It just works!The only qualm I have with it is the fact that it can only handle one minigel (or as the rep claimed, multiple strips of membrane equal to the area of one minigel). The primary antibody is also not recyclable. Overall, it's a neat little machine that does the "handsfree" antibody probing process very well.

Protocol Overview

The kit was used exactly as described by the manufacturer. An important point to note is that the primary and secondary antibodies were recommended to be used at 5 times the usual concentration, albeit in 2mL volume. So, supposing you normally use 10mLs of the diluted antibody, you'll be using the same quantity of conc. antibody.

Tips

I followed the manufacturer instructions to the letter and got a neat result. Although, I was using an antibody against a high expression protein, I'm not sure how it will do with low expression proteins

The Bands were clean, lower background than my usual (BSA, rolling tube) protocol.
