Target Enrichment in NGS

Whole-genome sequencing (WGS) technologies are evolving rapidly, reducing costs and shortening turnaround times. However, when the regions of interest are known ─ such as the exome, or specific gene groups ─ WGS isn’t always the most cost-effective route to sequencing results.

In these cases, targeted NGS provides a focused sequencing approach for select regions.  Compared to WGS, targeted NGS can:

  • enhance the detection of rare or low-frequency variants by yielding greater precision and deeper sequencing coverage;
  • accelerate the sequencing of more samples (throughput);
  • simplify data analysis;
  • reduce data storage needs…

all while consuming only a fraction of the per-sample sequencing resources required by WGS.

The NEW KAPA Target Enrichment portfolio from Roche offers a hybridization-based approach to capturing target genomic regions, enabling you to focus your sequencing resources on the genomic regions most relevant to your research.

KAPA Target Enrichment probes are the central components of the HyperCap Workflow v3.0, a rapid workflow that yields sequencing-ready target-enriched libraries in less than two days. This new workflow leverages:  high-precision KAPA Target Enrichment probes for either predesigned or custom targets;  the robust KAPA HyperPrep or KAPA HyperPlus Library Preparation Kits; the new KAPA UDI Primer Mix 1-96 (mixes 97-384 will be available soon); and KAPA Universal Enhancing Oligos.

The KAPA Target Enrichment portfolio includes panels for capturing the human exome (KAPA HyperExome) or customized genomic targets—KAPA HyperChoice for human targets, and KAPA HyperExplore for nonhuman targets. These custom panels are designed by customers using HyperDesign, Roche’s new user-friendly online target enrichment design software.
