Assay Principle
Gamma-aminobutyric acid (GABA) is the principal inhibitory neurotransmitter of the mammalian central nervous system, also studied for effects on blood pressure regulation, growth hormone secretion, and ethanol metabolism. A GABA assay generally relies on one of two distinct detection chemistries. Colorimetric kits, generally applied to plant and animal tissue homogenates, rely on a reaction between phenol, sodium hypochlorite, and GABA that produces a blue-green product with maximum absorbance around 640 nm. Fluorometric kits instead use an enzymatic reaction in which GABA is catalyzed with the removed hydrogen reducing NADP+ to NADPH, and GABA content is calculated from the resulting NADPH signal, read at approximately Ex/Em 535/587 nm.
