Description

Intended Uses: Human PAI-1 uPA complex assay is intended for the quantitative determination of the covalent complex of uPA and its inhibitor PAI-1 in biological fluids.

Principle of the Assay: Human uPA in samples will bind to the capture antibody coated on the microtiter plate. Free and complexed enzyme will react with the capture antibody on the plate. After appropriate washing steps, polyclonal anti-human PAI-1 primary antibody binds to PAI-1 uPA complex captured on the plate. Excess antibody is washed away and bound polyclonal antibody is then reacted with the secondary antibody conjugated to horseradish peroxidase. TMB substrate is used for color development at 450nm. A standard calibration curve is prepared along with the samples to be measured using dilutions of CPAI uPA complex. Color development is proportional to the concentration of PAI-1 uPA complex in the samples. Free uPA and PAI-1 will not be detected by this assay.
