Description

Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with Nitrofurazone antigen. Standards or samples are added to the appropriate microtiter plate wells with Nitrofurazone specific antibody and Horseradish Peroxidase (HRP) conjugated anti-antibody. The competitive inhibition reaction is launched between pre-coated Nitrofurazone and Nitrofurazone in standards or samples with the Nitrofurazone special antibody. A substrate solution is added to the wells and the color develops in opposite to the amount of Nitrofurazone in the standards or samples. The color development is stopped and the intensity of the color is measured.

Background/Introduction: Nitrofurans are synthetic broad-spectrum antibiotics, widely used in animal production for its excellent antibacterial and pharmacokinetic properties. They had been also used as growth promoters in the pig production, the poultry and fish sector. In long term studies with experimental animals the parent drugs and their metabolites showed carcinogenic and mutagenic characteristics. This has led to a prohibition of Nitrofurans for the treatment of animals used for food production. Current methods of detection Nitrofurazone, such as LC-UV, LC-MS and LC-MS/MS, are with sample preparation complex, expensive, difficult and time-consuming.
