Introduction: 5-Hydroxytryptamine, also known as serotonin is a monoamine neurotransmitter. It is found extensively in the gastrointestinal tract of animals, and about 80 to 90 percent of the human body's total serotonin is located in the enterochromaffin cells in the gut, where it is used to regulate intestinal movements. The remainder is synthesized in serotonergic neurons in the central nervous system (CNS) where it has various functions, including the regulation of mood, appetite, sleep, muscle contraction, and some cognitive functions including memory and learning.
Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with an goat-anti-rabbit antibody. Standards or samples are added to the appropriate microtiter plate wells with an antibody specific for 5-HT and HRP-conjugated 5-HT, then incubated. A competitive inhibition reaction is launched between 5-HT (Standards or samples) and HRP- conjugated 5-HT with the antibody specific for 5-HT. The more the amount of 5-HT in samples, the less antibody bound by HRP)-conjugated 5-HT. The substrate solutions are added to the wells, respectively. And the color develops in opposite to the 3 amount of 5-HT in the sample. The color development is stopped and the intensity of the color is measured
