Introduction: Clara Cell Protein belongs to the family of secretoglobins. It is a secreted protein product of non-ciliated bronchiolar Clara cells. It is the founding member of the secretoglobin family of small, secreted, disulfide-bridged dimeric proteins found only in mammals.This antiparallel disulfide linked homodimeric protein is multifunctional and found in various tissues in various names such as: Uteroglobin (UG, UGB), Uteroglobin-like Antigen (UGL), Blastokinin, Clara cell secretory protein (CCSP), Clara cell 16 kD protein (17 in rat/mice), Clara Cell-Specific 10 kD Protein (CC10), human protein 1, urine protein 1 (UP-1), polychlorinated biphenyl-binding protein (PCB-BP), human Clara Cell Phospholipid-Binding Protein (hCCPBP), Secretoglobin 1A member 1 (SCGB1A1).
Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to CC16. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for CC16 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain CC16, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of CC16 in the samples is then determined by comparing the O.D. of the samples to the standard curve
