Western blot was performed using Anti-KCNN3 Polyclonal Antibody (Product # PA5-41014) and a 90 kDa band corresponding to KCNN3 was observed in the Mouse and Rat Brain tissues where as no band was observed in Cerebellum and Spleen which are very low expression models for KCNN3. Tissue extracts (100 µg lysate) of Mouse Brain (Lane 1), Rat Brain (Lane 2), Mouse Cerebellum (Lane 3), Mouse Spleen (Lane 4) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:500) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036,1:10000) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Atto Ultimate Sensitivity Substrate (Product # A38556). * indicates a non specific band observed in Rat Brain.
