Western blot was performed using Sortilin Polyclonal Antibody (Product # PA5-47462) and a 110kDa band corresponding to SORT1 was observed across all cell lines and tissues tested except in Mouse and Rat liver. Whole cell extracts (30 µg lysate) of SK-BR-3 (Lane 1), MDA-MB-231 (Lane 2), BeWo (Lane 3), HeLa (Lane 4), Mouse brain (Lane 5), Rat brain (Lane 6), Mouse liver (Lane 7), Rat liver (Lane 8) were electrophoresed using NuPAGE® 4-12 % Bis-Tris gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Rabbit anti-Goat IgG Heavy Chain Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27014, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
