(A, B) Mammosphere formation assay was performed using SUM159PT mammospheres plated in a 96-well plate and treated with different concentrations of TNFα (10, 50, 100, 500 ng/mL) or vehicle (0.1% BSA). The number of mammospheres formed in each condition was counted and normalized against the vehicle control. (C) The SUM159PT cells were plated at a density of 200 cells per well in a 6-well plate, then treated with a single dose of TNFα (1, 10, 50, 100 ng/mL) or vehicle (0.1% BSA) and cultured for 14 d.
