Description
Axl, also known as UFO, Tyro 7 and ARK, is a receptor tyrosine kinase, is involved in cell survival, proliferation, and migration and have sequence homology with other 2 members of the receptor tyrosine kinase family, MER and Tyro3. This tyrosine kinase is activated by the Growth Arrest Gene 6 (GAS 6) that binds and activated the kinase domin of this receptor kinase. Based on sequence homology and secondary structure predictions, AXL putative protein has a tyrosine kinase domain with unique 2 immunoglobulin like domain in the C-terminal end. It also has 2 fibronectin type III domains which are found on proteins such as receptor tyrosine phosphatases, and cell adhesion molecules. The expression of mRNA for this gene was noted in broad spectrum of developing tissues (e.g. subepidermal cells of the skin, mesenchymal cells of the periosteum). AXL expression is also detected in most cell lines and adult tissues except those of the hmatopoietic lineage (1, 2). The receptor Axl is expressed in primary prostate carcinoma and in prostate cancer cell lines as such as PC-3 and DU 145. Axl expression increases in the neointima of balloon-injured rat carotids due to the oxidative stress which also plays a role in remodeling of injured vessels. Several cancer cell lines express Axl protein including primary prostate carcinoma and in prostate cancer cell lines as such as PC-3 and DU 145, myeloid leukemia. GAS 6 activation of Axl is also invloved in experimental nephritis model and spermatogenesis (3). IN testis, AXL is expressed in sertoli and leydig cell of the mature testis. The receptor tyrosine kinase AXL protein has has at least 2 variants (AXL1, AXL 2) and is a approximately 130 kDa (888 amino acids) protein. The AXL protein has simlar functional domins as its other family members and has a tyrosine kinase domain closer to the C-terminal region and putative phosphorylation sites. The Anti-AXL-selective antibody was generated against a conserved sequence near the C-terminal that is unique to AXL protein only. The polyclonal antibody strongly labels a 130kDa protein in Western bl;otpositive control sample for AXL (PC-AXL). Anti-AXL-selective antibody is also available in affinity-purified form for confocal, Western blotting and immunocytochemical analyses
