The PDE4 family of cAMP-specific phosphodiesterases represents an important therapeutic and pharmacological target for a variety of therpapeutic interventions including their anti-inflammatory and anti-depressant effects. Four distinct genes (PDE4A, PDE4B, PDE4C and PDE4D), encode large family of PDE4 isoforms [1, 2). Almost all of these isoforms have a common core made up of central catalytic domain and C-terminal tail. The differential RNA splicing and use of alternate initiation codons also generates the elongated N-terminal ends "long-forms" PDE4 proteins. The long form PDE variants are characterized by upstream conserved regions (UCR1 and UCR2). The PDE4A5 is a rat/mouse homologue of human PDE4A4 long-form PDE4A variant. The PDE4A5 has the highest sensitivity to rolipram inhibition (3nM) that is at least 100-fold less than for other PDE4 family members in U937 cells (3). PDE4A5-selective antibody was generated against unique N-terminal sequence, the peptide was post-synthetically modified to achieve maximum antigeneicty and provide proper linkers for covalent coupling to carrier protien (blue carrier protien). The anti- PDE4A5 antibodies does not cross-react with other PDE4A proteins including A4, A1 and Ax.
