Western blot was performed using Anti-TDP-43 Polyclonal Antibody(Product # PA1-16996) and a 44 kDa band corresponding to TDP-43 was observed across cell lines and tissue extracts tested. A non-specific band around 110 kDa has also been observed in HeLa, MCF-7 and Caco-2. Whole Cell Extract-WCL (30 µg lysate) of HeLa (Lane 1), MCF7 (Lane 2), Caco-2 (Lane 3), A549 (Lane 4), K-562 (Lane 5), Neuro-2a (Lane 6), NIH/3T3 (Lane 7), PC-12 (Lane 8) and tissue extracts of Mouse Brain (Lane 9), Rat Brain (Lane 10) and Mouse Thymus (Lane 11) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (0.5 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4,000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
