Western blot was performed using Anti-MMP2 Monoclonal Antibody (4D1E2) (Product # MA5-17122) and a 70kDa band corresponding to MMP2 was observed. Whole cell extracts (40 µg lysate) of U-87 MG (Lane 1), HT-1080 (Lane 2), LNCaP (Lane 3) and tissue extract (40 µg lysate) of Mouse Uterus (Lane 4) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005). Band of interest was not observed in LNCaP, a cell line that expresses very low levels of MMP2 (https://doi.org/10.1210/en.2002-0157).
