Western blot was performed using Anti-LGALS4 Polyclonal Antibody (Product # PA5-32251) and a 36 kDa band corresponding to LGALS4 was observed in HT-29 and COLO 205 but not in HCT 116, Caco-2, HeLa and A549. Whole cell extracts (30 µg lysate) of HT-29 (Lane 1) HCT 116 (Lane 2), Caco-2 (Lane 3), COLO 205 (Lane 4), HeLa (Lane 5) and A549 (Lane 6) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1,000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).* - There was an uncharacterized band observed at ~28kDa in Caco-2.
