Western blot was performed using Anti-MSH6 Polyclonal Antibody (Product # PA5-29348) and a 160 kDa band corresponding to MSH6 was observed in all the tested cell models and tissue lysates, except Jurkat and LNCaP which are reported to be negative. This target undergoes heavy phosphorylation and multiple bands between 120-160 kDa can be expected. Modified whole cell lysate (1% SDS) (30 µg lysate) of SH-SY5Y (Lane 1), K-562 (Lane 2), Jurkat (Lane 3), HeLa (Lane 4), LNCaP (Lane 5), PC-3 (Lane 6), HCT 116 (Lane 7), A549 (Lane 8) and tissue extract (30 µg lysate) of Mouse Testis (Lane 9) were electrophoresed using NuPAGE® 10 % Bis-Tris gel (Product # NP0302BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1,000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4,000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
