Western blot was performed using Anti-CNPase Polyclonal Antibody (Product # PA5-27972) and a 47 kDa band corresponding to CNPase was observed in Mouse brain and Mouse cerebellum when compared to Mouse Kidney, Mouse Lung and Mouse Colon which are reported to be low. CNPase is expressed exclusively by oligodendrocytes in the central nervous system. Membrane enriched extracts (30 µg lysate) of MOLT-4 (Lane 1), Hep G2 (Lane 2), SK-BR-3 (Lane 3), HeLa (Lane 4), tissue extracts of Mouse Brain (Lane 5), Mouse Cerebellum (Lane 6), Mouse Kidney (Lane 7), Mouse Lung (Lane 8) and Mouse Colon (Lane 9) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:5000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
