Western blot was performed using Anti-beta-2 Microglobulin Polyclonal Antibody (Product # PA5-29580) and a 12 kDa band corresponding to beta-2 Microglobulin was observed across in HeLa, THP-1 and U-937 but not in SH-SY5Y or NTERA-2 cell lines which are reported to express low levels of beta-2 microglobulin. Whole cell extracts (60 µg lysate) of HeLa (Lane 1), THP-1 (Lane 2), U-937 (Lane 3), SH-SY5Y (Lane 4) and NTERA-2 cl.D1 (Lane 5) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:500) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036,1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
