Pyruvate dehydrogenase complex (PDC) catalyzes the oxidative decarboxylation of pyruvate and links glycolysis to the tricarboxylic acid cycle and fatty acid synthesis. PDPR is the regulatory subunit of mitochondrial pyruvate dehydrogenase phosphatase (EC 3.1.3.43). It inhibits the Mg(2+)-dependent and Ca(2+)-stimulated activity of the catalytic subunit (PDP1; 605993) by decreasing its sensitivity to Mg(2+). PDPR decreases the sensitivity of PDP1 to magnesium ions, and this inhibition is reversed by the polyamine spermine.
