Western blot was performed using Anti-Cullin 3 Polyclonal Antibody (Product # PA5-17397) and a 81 kDa band corresponding to CUL3 was observed across cell lines and tissues tested. (Fig. a) Whole cell extracts (30ug) of HeLa (Lane 1), PC-12 (Lane 2), Neuro-2a (Lane 3), L6 (Lane 4), U-2 OS (Lane 5), Hep G2 (Lane 6), MDA-MB-231 (Lane 7), MCF 10A (Lane 8) and T-47D (Lane 9), (Fig.b) tissue extracts (30ug) of Mouse Brain (Lane 1), Rat Liver (Lane 2), Mouse Liver (Lane 3), Rat Ovary (Lane 4) and Mouse Ovary (Lane 5) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat Anti-Rabbit IgG Secondary Antibody, HRP conjugate (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
