Western blot was performed using anti-AGO2 Monoclonal Antibody (R.386.2) (Product # MA5-14861) and a band at 97 kDa corresponding to AGO2 was observed across the cell lines tested and was increased upon LPS treatment in THP-1. An uncharacterized band (*) was also observed at ~20 kDa. Modified whole cell extracts (1% SDS) (30 µg lysate) of MCF7 (Lane 1), MDA-MB-231 (Lane 2), T-47D (Lane 3), Hep G2 (Lane 4), THP-1 (Lane 5), THP-1 treated with LPS (1 µg/mL for 24hrs) (Lane 6) and RAW 264.7 (Lane 7) were electrophoresed using Novex® NuPAGE® 4-12% % Bis-Tris gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1,000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4,000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
