Western Blot was performed using Anti-PDGF-B Polyclonal Antibody (Product # PA5-19524) and a 17 kDa band corresponding to Platelet-derived growth factor subunit B was observed across cell lines tested and was upregulated in cell lines upon PTI treatment. Also, the expression was found to be high in HUVEC when compared to SH-SH5Y. Whole cell extracts (30 µg lysate) of HMEC-1 (Lane 1), HUVEC (Lane 2), HUVEC treated with PTI (1X for 4 hrs) (Lane 3), SH-SY5Y (Lane 4), SH-SY5Y treated with PTI (1X for 4 hrs) (Lane 5) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0301BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:6,000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).
