Immunofluorescent analysis of HeLa cells fixed in 3.5% paraformaldehyde using Product # PA3-16832 (1:1,000). Nuclear staining during interphase and spindle staining during mitosis.

Western blot was performed using Anti-TPX2 Polyclonal Antibody (Product # PA3-16832) and a ~110 kDa band corresponding to targeting protein was observed along with an uncharacterised band (*) at ~40 kDa across cell lines tested. Nuclear enriched extracts (40 µg lysate) of HeLa (Lane 1), A431 (Lane 2), A549 (Lane 3), MCF7(Lane 4), HCT116 (Lane 5) and NIH/3T3 (Lane 6) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036,1:20000 dilution) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).
