VAMP8 (also known as endobrevin), a member of the soluble N-ethylmaleimide-sensitive fusion protein attachment protein receptor (SNARE) family of fusion proteins, initially characterized in endosomal and endosomal-lysosomal fusion, may also function in regulated exocytosis. VAMP8 gene plays an important role in biological functions like endosomal fusion, sequential granule-to-granule fusion, and autophagy. The VAMP8 vesicle-associated-SNARE is required for the proper trafficking of secretory lysosomal granules for exocytosis in macrophages and the release of the potent pro-inflammatory cytokine, TNF-alpha. The VAMP8 acts as a novel oncogene by promoting cell proliferation and therapeutic resistance in glioma. VAMP8 is significantly upregulated in human breast cancer (BC) tissues and can thus be a practical and potentially effective surrogate marker for survival in BC patients
