AlkB homolog 5 (ALKBH5), an m(6)A demethylase, specifically removes m6A from target mRNAs. ALKBH5, which is most highly expressed in the testes but has low expression in the heart and brain, affects nuclear RNA export and metabolism, gene expression and mouse fertility. Alkbh5 knockout mice were viable but showed compromised spermatogenesis due to significantly altered expression of key genes required for spermatogenic maturation although the increase of overall m6A levels in testes is modest. ALKBH5-mediated m6A erasure in the nuclei of spermatocytes and round spermatids is essential for correct splicing and the production of longer 3'-UTR mRNAs, and failure to do so leads to aberrant splicing and production of shorter transcripts with elevated levels of m6A that are rapidly degraded.
