Western Blotting Analysis of HeLa acid extract was probed with 029502 (1ug/ml). Proteins were visualized using a IgG (HRP) Donkey Rabbit and a chemiluminescence detection system. Arrow indicates Histone H3.3 (~17kD).

Immunofluorescence analysis of HeLa cells, stably transfected with Flag-HA- tagged H3.3. Cells were fixed and permeabilized with 4% paraformaldehyde in PBS-T (PBS/0.1% Triton X-100) for 10min at RT. Cells were then incubated with 029502 (2ug/ml) in PBS-T overnight at 4°C and subsequent fluorescent secondary antibody detection under a confocal microscope.
