GAP43 (also known as neuromodulin)is considereda "growth" or "plasticity" protein because it is expressed at high levels in neuronal growth cones during developmentand axonal regeneration, and it is phosphorylated after long-term potentiation and after learning.It isanervous tissue-specificproteinthat localizes to the cytoplasm of neurons and attaches to the membrane via a dual palmitoylation sequence on cysteines 3 and 4. GAP-43is a major protein kinase C (PKC) substrate. A nullmutationof GAP-43leads to death within days after birth, due to axon pathfinding defects. Additionally, aberrant expression of GAP43 has been seen in patients identified with schizophrenia and Alzheimer's disease
