HCoV-HKU1 is the one of seven known coronaviruses to infect humans, including HCoV-229E, HCoV-NL63, HCoV-OC43, MERS-CoV, the original SARS-CoV (or SARS-CoV-1), and SARS-CoV-2.
The virus is an enveloped, positive-sense, single-stranded 29,926-nucleotide polyadenylated RNA virus which enters its host cell by binding to the N-acetyl-9-O-acetylneuraminic acid receptor. It has the Hemagglutinin esterase (HE) gene, which distinguishes it as a member of the genus Betacoronavirus and subgenus Embecovirus.
The genome organization is the same as that of other Betacoronaviruses, with the characteristic gene order 1a, 1b, HE, S, E, M, and N. Furthermore, accessory protein genes are present between the S and E genes (ORF4) and at the position of the N gene (ORF8)
