Immunofluorescence analysis of SELP was performed using log phase HEL 92.1.7 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 45 minutes, and blocked with 2% BSA for 1 hour at room temperature. The cells were labeled with P-Selectin Recombinant Rabbit Monoclonal Antibody (JM10-95) (Product # MA5-41223) at 1:100 dilution in 0.1% BSA, incubated at 4 degree celsius overnight and then labeled with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034), (1:2000 dilution), for 45 minutes at room temperature (Panel a: Green). Nuclei (Panel b:Blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: Red) was stained with Rhodamine Phalloidin (Product # R415, 1:300 dilution). Panel d represents the merged image showing cytoplasm (plasma membrane like) localization. Panel e represents Raji cells which is a negative model for SELP. Panel f represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.
IHC (P)

Immunohistochemistry (Paraffin) analysis of P-Selectin-CD62P- in paraffin-embedded human spleen tissue using P-Selectin Recombinant Monoclonal Antibody (Product # MA5-41223). Counter stained with hematoxylin.

Fig a- Western blot was performed using Anti-P-Selectin Recombinant Rabbit Monoclonal Antibody (JM10-95) (Product # MA5-41223) and a 140 kDa band corresponding to SELP, along with an uncharacterized band (*), was observed across positive cell line (HEL 92.1.7) and not negative cell line (Raji). Whole cell extracts (40 µg lysate) of HEL 92.1.7 (Lane 1), Raji (Lane 2) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1500 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:20000 dilution) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Atto Ultimate Sensitivity Substrate (Product # A38556). Fig b- The same primary antibody was tested in Huvec (Lane 1) and Huvec treated with IL-4 (20 ng/mL for 24 hours) (Lane 2) and a 140 kDa band corresponding to SELP was observed along with an uncharacterized band (*).
