Sodium-phosphate cotransporter 2a, or NaPi2a (SLC34A1), is a solute-carrier (SLC) transporter located in the kidney proximal tubule that reabsorbs glomerular-filtered phosphate. Slc34a1 resides downstream of the gene encoding coagulation factor XII (f12). The Na+-dependent phosphate transporter NaPi-IIa (SLC34A1) is mostly expressed in the kidney, whereas NaPi-IIb (SLC34A2) has a wider tissue distribution with prominent expression in the lung and small intestine. Inhibition of NaPi2a may enhance urinary phosphate excretion and correct maladaptive mineral and hormonal derangements associated with increased cardiovascular risk in chronic kidney disease-mineral and bone disorder (CKD-MBD). Mutations in SLC34A1 may cause a range of clinical phenotypes including and idiopathic infantile hypercalcemia and nephrocalcinosis, a proximal renal Fanconi syndrome, which are typically autosomal recessive, and hypophosphatemic nephrolithiasis, which may be an autosomal dominant trait
