The PPP2R1A gene on chromosome 19 (19q13.41) encodes the alpha isoform of the scaffolding subunit of the PP2A holoenzyme, which functions to link the catalytic subunit to the regulatory subunit. Somatic missense mutations in the Ser/Thr protein phosphatase 2A (PP2A) Aalpha scaffold subunit gene PPP2R1A are among the few genomic alterations that occur frequently in serous endometrial carcinoma (EC) and carcinosarcoma, two clinically aggressive subtypes of uterine cancer with few therapeutic options. The PPP2R1A mutation regions are conserved among species and known to interact with the regulatory subunits of the PP2A enzyme. Subtype-specific PPP2R1A mutations have been frequently observed in ovarian and endometrial cancer. PPP2R1A mutant endometrial cancers may represent good candidates for personalized drug therapies particularly for women with the lethal serous histologic variant of uterine cancer
