DiGeorge syndrome chromosomal region 8 (DGCR8), a double-stranded-RNA-binding protein, participates in the miRNA biogenesis pathway and contributes to miRNA maturation by interacting with the RNAase III enzyme Drosha in cell nuclei. DGCR8, which is known as Pasha in flies and worms, is a 773-amino-acid (aa) protein critical for microRNA (miRNA) biogenesis via the formation of a microprocessor complex with Drosha. This complex processes primary miRNA in the nucleus. DGCR8 deletion in murine B-cell progenitors impairs the transition from pro-to early pre-B cell stage, recapitulating the phenotypes observed in miR17-92 cluster knockout mice.
