JCAD (Junctional protein associated with Coronary Artery Disease; previously KIAA1462), located on chromosome 10, encodes 1359-amino-acid protein. JCAD is an intrinsically disordered protein with no recognizable functional domains and little homology to other protein families. JCAD has been identified as a novel component of endothelial cell junctions where it is found to co-localize with VE-Cadherin. A role for JCAD has been identified in pathological angiogenesis with decrease vascular formation in response to matrigel and decreased tumour growth. JCAD is an interactor in the Hippo pathway, and as a negative regulator of LATS2 kinase activity. JCAD negatively regulates Hippo signaling in endothelial cells. JCAD contributes to atherosclerosis by mediating YAP activity and contributing to endothelial dysfunction
