Rab3B, a member of the small GTP-binding protein Rab family, is known to be involved in Ca(2+)-regulated exocytosis in pituitary cells. Rab3B is a monomeric GTPase that modulates norepinephrine secretion when expressed in PC12 neuroendocrine cells. Rab3B is essential for GnRH-regulated exocytosis downstream of cytosolic Ca2+ in gonadotrophs. Rab3B can influence junctional protein targeting and secretion by distinct mechanisms. Rab3B, similar to other Rab3 isoforms, is a synaptic vesicle protein that interacts with the Rab3-interacting molecule (RIM) isoforms RIM1alpha and RIM2alpha as effector proteins in a GTP-dependent manner. RAB3B localized to the cytoplasm of human beta-cells, both during the fetal period and postnatally. The regulation of insulin exocytosis in rodent beta-cells and responsiveness to incretins are reliant on Rab family members, notably Rab3a and Rab27a, but not Rab3b
