T4 DNA Ligase is an ATP-dependent recombinant enzyme isolated from Escherichia coli strain used to clone DNA. T4 DNA Ligase catalyzes the formation of a phosphodiester bond between the terminal 5' phosphate and 3' hydroxyl groups of duplex DNA or RNA. The enzyme efficiently joins blunt and cohesive ends and repairs single-stranded nicks in duplex DNA, RNA, or DNA/RNA hybrids. It can also ligate blunt-ended DNA with much greater efficiency than E Coli DNA ligase. Unlike E Coli DNA ligase, T4 DNA ligase cannot utilize nicotinamide adenine dinucleotide (NAD) and it has an absolute requirement for ATP as a cofactor.
