Members of the NEK (NIMA-related kinase) family of serine/threonine protein kinases have been implicated in the regulation of various aspects of the cell cycle. NEK10 structure is unique in that it has a central catalytic kinase domain, flanked by two large regulatory domains. Like NIMA and NEKs 1, 2, 5, 9, and 11, the NEK10 protein has a coiled-coil region, located near the kinase domain. In the amino-terminal regulatory domain there are four armadillo motifs, which also act as key regions for protein-protein interactions. Mutations in NEK10 have been reported in lung cancer and breast cancer, in which polymorphisms in BRCA1/2 (breast cancer type 1/2 susceptibility protein) were found. NEK10 is important for ciliogenesis. NEK10 interacts with PKA and PCM1 and participates in a cAMP dependent pathway, contributing to cilium formation.
