Description

One of the proteins involved in modifying ribosomes so they are functional is called BUD23. BUD23 in Saccharomyces cerevisiae encodes for a class I methyltransferase, and deletion of the gene results in slow growth and random budding phenotypes. Deletion of BUD23 leads to reduced levels of the small (40S) ribosomal subunit, and a block in processing 20S rRNA to 18S rRNA, a late step in 40S maturation. Bud23 protein, but not its methyltransferase activity, is important for biogenesis and export of the 40S subunit in yeast. Baxter et al. found that ribosomes in human cells with no BUD23 were different than in normal cells and that cells without BUD23 produced different proteins, which did not always perform their roles correctly. Proteins in the mitochondria are one of the main groups affected by the absence of BUD23
