TRAPP complex, a multi-subunit tethering complex and GEF for RAB1, is an interactor of TBC1D14. TBC1D14 binds to the TRAPP complex via an N-terminal 103 amino acid region, and overexpression of this region inhibits both autophagy and secretory traffic. TBC1D14 has a typical domain structure for TBC domain-containing RabGAPs, with the TBC domain at the C-terminus of the protein. TRAPPC8 is required for TBC1D14 to bind the TRAPP complex. TBC1D14 may act to balance direct, bulk contribution of RE to autophagosomes through its RAB11-binding ability, while promoting constitutive ATG9 cycling from the Atg9 compartment through its recruitment of TRAPP and activation of RAB1
